It's simply the number of layers of insulating film that have been applied 
to the wire. The heavier the "build", the thicker and tougher the 
insulation layer, so it's harder to nick during/after winding. Multiple 
builds will also give you a higher turn-to-turn breakdown voltage - 
important if you plan to push your secondary to the limit. I've always used 
at least double build with excellent results.

BTW, a source for just about any type of wire is Joe Cassata at the Coax 
Connection, KA9CAI. He stocks, or can get, just about any gauge and type of 
wire you're looking for, and he's my supplier for all of my magnet wire 
needs (and I go though a LOT when shrinking coins!).
